The Jogoos have only beaten relegation candidates Soltilo Bright Stars away from home and sit 8th on the log with 33 points while Mbarara City are 4th from bottom with only 23 points.
Seven-time champions Express face Mbale Heroes at the Municipal stadium looking to get back to winning ways after five games on the trot with a win.
They will hope to build on the 2-2 draw with table leaders Vipers last week, a result that saw the title race wide open again.
Mbale Heroes are bottom of the table with only 10 points at this stage and are a side everyone wants to play at the moment although they can be a nuisance at their own home.
KCCA will make the shortest trip of the trio as they travel to Nakivubo to take on four-time winners URA in a late kick off under floodlights.

KCCA are in 6th position while URA, who they beat 2-0 in the reverse fixture at Lugogo on 39 points in fourth place.
Elsewhere, BUL will be desperate to extend their unbeaten streak in the second round when they host Police at Njeru.
Both teams come off good victories with Police beating Mbarara City 2-0 while Bul saw off KCCA 2-1 at Lugogo to keep 3rd position on 42 points and understand that a win at Njeru will see them trim the gap at the top.
In the other match on Wednesday, Maroons visit UPDF at Bombo in a game between security forces.
The army side on 29 points in 10th place are yet to mathematically survive the drop and will be eager for a win while Maroons also want to win after a disappointing 3-3 draw with Wakiso Giants in the last outing despite leading 3-1 at one moment in the second half.
