Vipers must win at Mbarara City to keep their slim chances of retaining the title when the two sides face off at Kakyeeka stadium on Wednesday.
The champions head into the game on the back of two winless games but face a tough test in Mbarara City who are seeking a return to winning ways.
The corresponding fixture last season ended in a 1-0 win for the Venoms thanks to controversy after Clovis Mbayi goal tore the net.
Daniel Sserunkuma could return to the starting eleven against a side that will have defender Hilary Mukundane return from suspension.
The hosts are expected to be without Paul Mucureezi (injury) and goalie Ali Kimera (internal suspension) for the game.
The reverse fixture at Kitende ended in a 1-1 draw but the Ankole Lions have never won against the Venoms.
Elsewhere, URA host Bright Stars at Namboole seeking to avenge the Uganda Cup defeat, Tooro United could go fourth if they win away to struggling Maroons while in Jinja, there is a derby that pits Kirinya against Bul at Njeru.
